i did
> set G4OPTDEBUG=1
> export G4OPTDEBUG
> ./Configure -build -D G4OPTDEBUG

but I'm not sure i set this flag properly. I checked the logs and scripts in .config and I cannot find there "-O" of G4OPTDEBUG.

How do I know that Configure understands my intention to build using G4OPTDEBUG? It's building now, but I'm afraid it is just a simple unoptimized debug build. (I answered yes to DEBUG question in Configure script).
